摘  要:目的:优选中药麦芽发芽炮制工艺。方法:以麦芽中总生物碱含量、大麦芽碱含量及淀粉酶活力为指标,在单因素试验基础上,通过正交试验考察浸泡时间、发芽温度、发芽湿度及每日洒水量对大麦发芽的影响。依次采用酸性染料比色法和高效液相色谱法测定总生物碱与大麦芽碱含量,DNS比色法测定淀粉酶活力。结果:优选出的麦芽最佳发芽工艺为:用水浸泡5h,发芽温度25℃,湿度70%,每日洒水量与大麦重量比为1∶1。结论:优选得到的炮制工艺稳定可行。OBJECTIVE To optimize the germination processing technology of the traditional Chinese medicine malt. METItODS The contents of total alkaloids, hordenine in malt and amylase activity were used as index. Based on the single factor experiment, the effects of soaking time, germination temperature, germination humidity and daily watering amount on the barley germination were investigated by orthogonal test. The contents of total alkaloids and hordenine acid were determined by dye colorimetry and HPLC, amylase activity was determined by DNS colorimetry. RESULTS The optimized malt best ger- mination process: soaking time for 5 h, germination temperature at 25 ℃, humidity at 70%, the weight ratio of daily watering amount and barley was 1 : 1. CONCLUSION The optimized processing technology is stable and feasible.
