补阳还五汤对脑缺血后3天和15天miRNA表达的影响

摘　　要：目的探讨补阳还五汤对大脑中动脉栓塞后不同时间大鼠脑内微小核糖核酸(miRNA)表达的影响。方法建立大脑中动脉栓塞的脑缺血模型,芯片方法观察miRNA的表达差异,RT-PCR进行验证。结果与3天模型组相比,补阳还五汤组miRNA有2个上调,1个下调;与15天模型组相比,补阳还五汤组miRNA有5个上调,4个下调。随后,选取3个miRNA使用RT-PCR验证各组大鼠脑内miRNA差异表达,同芯片结果相符。结论补阳还五汤对脑缺血后3天和15天miRNA均能产生影响,提示补阳还五汤对脑缺血急性期和修复期都具有治疗作用。另外,相比较而言,补阳还五汤对脑缺血后15天miRNA影响的数量更多,可能其促进修复的作用更具有优势。Objective To investigate the effects of Buyang Huanwu Decoction（ BYHWD） on miRNA after focal ischemia at different duration. Methods Middle cerebral artery occlusion and reperfusion（ MCAO/R） was used as the model of focal ischemia,microarray was used to observe their expression levels,and real-time polymerase chain reaction（ RT-PCR） was used to verify the data. Results Compared with ischemia day 3 group,BYHWD up-regulated 2 miRNAs and down-regulated 1; Compared with ischemia day 15 group,BYHWD up-regulated 5 miRNAs and down-regulated 4. Then 3 miRNAs expression were evaluated by RT-PCR,which was consistent with microarray data. Conclusion BYHWD have effects on miRNAs at both day 3 and day 15,indicating that ischemia could be treated by BYHWD during both acute and reparing stage. Notably,BYHWD can influence more miRNAs on day 15 than that on day 3,indicating BYHWD might have advantage to promote repairment after ischemia.
