摘 要:研究了分批培养、pH反馈补料培养、定量补料培养与培养基完全更换培养4种培养模式对雨生红球藻绿色生长以及虾青素积累的影响。结果表明:当在绿色细胞阶段采用培养基完全更换的培养模式,能够有效的促进雨生红球藻细胞的分裂,同时藻细胞保持了较高活性。其中培养9d后藻细胞数目达到8.60E+05个/mL,游动细胞所占比例达到98.26%,细胞干重为1.12g/L,而且胁迫条件下培养10d后虾青素的产量达到53.21mg/L,明显高于其他培养模式。因此培养基完全更换培养是一种适合雨生红球藻生长与积累虾青素的培养模式。The effect of four kinds of culture modes (batch culture, quantitative fed-batch, fed-batch based on pH and total replacement of medium) on green growth and astaxanthin accumulation by Haematococcus pluvialis was investigated. The results showed that total replacement of medium promoted cell division effectively. At the same time, the proportion of motile cells reached 98.26 %, indicating high bioactivity of algal cells. The numbers of algal cells reached 8. 60E+05 cells/mL after 9 days, while the cell dry weight was 1.12 g/L and astaxanthin production was 53.21 mg/L, which were significantly higher than the other three culture modes. Therefore, total replacement of medium is a suitable culture mode for algal cells growth and astaxanthin accumulation in Haernatococcus pluvialis.
