摘  要:试验以驼血为原料,利用响应曲面法(RSM)对驼血多肽的制备工艺进行优化。以酶解产物中低聚肽所占比例为响应值,考察蛋白酶A的加入量、酶解时间和酶解温度3个因素对驼血多肽酶解工艺的影响。在单因素试验的基础上,选取试验因素与水平,根据中心组合(Box-Behnken)试验设计原理,采用三因素三水平的响应曲面法对各个因素的显著性和交互作用进行分析。结果表明:驼血多肽酶解的最佳工艺条件为:蛋白酶A的加入量2.47%,酶解时间3 h,酶解温度60℃,该条件下酶解液中低聚肽所占比例为98.51%。试验可为从驼血中分离制备驼血多肽提供技术支持。The preparation process of polypeptides from camel blood was optimized by response surface methodology using camelblood as raw material in this study. Using the ratio of oligopeptides in the hydrolysate as response value, the effect on hydrolysis process was studied through the investigation of amount of protease A, hydrolysis time and hydrolysis temperature. The one-factor-at-a-time experiments were carried out to figure out the effects of three parameters on the preparation of polypeptides from camel blood. According to these results, the experiment factors and its levels were selected. And significance and interaction of each factor were analyzed on the basis of a three-variable, three-level central composite design based on Box-Behnken model. From the above investigations and analyses, optimum process conditions were determined to be: amount of protease A of 2.47%, hydrolysis time of 3 h and temperature of 60 ℃. Under the optimum conditions, from the view of theoretical calculation, ratio of oligopeptides in the hydrolysates was 98.51%. The results would provide the technical support for the preparation of polypeptides from camel blood.
