摘 要:为优选金针菇中总多酚含量测定的方法和条件,以方法学验证结果等作为评价指标,考察福林酚法、紫外分光光度法及普鲁士蓝法测定金针菇多酚含量的适用性,选择最佳测定方法;再以吸光度为响应值,对优选方法显色条件进行单因素考察,在此基础上采用二次正交旋转组合设计进行优化。结果表明:综合比较3种方法干扰性、灵敏度及稳定性等,福林酚法更适用于金针菇总多酚含量的测定;条件优化后最佳显色参数为福林酚显色剂2 mL,15%Na_2CO_3溶液2.4 mL,时间70 min,温度45℃,且该方法条件优化结果拟合度高,稳定可靠,可大大提高测定金针菇总多酚的灵敏度。To optimize the conditions to determinate the total polyphenol in Flammulina velutipes with validation results and other evaluating indicators,the applicability of Folin-Ciocalteu method,UV spectrophotometry and Prussian blue method was investigated,and the method for determination of total polyphenol in Flammulina velutipes was selected.With the absorbance as indicator,single-factor experiments of chromogenic conditions were studied,and the chromogenic conditions were optimized by quadratic orthogonal rotation combination design.Results showed that comparing the interference,sensitivity and stability of the three methods,Folin-Ciocalteu method was the optimal one for the determination of total polyphenol in Flammulina velutipes.The optimum chromogenic conditions were 2 mL Folin-Ciocalteu reagent,2.4 mL 15% Na_2CO_3 at 45 ℃ for 70 min.The method was accurate,stable and reliable for the determination of total polyphenol in Flammulina velutipes with greatly improved detection sensitivity.
