风心病合并房颤患者心肌microRNA表达谱分析与靶基因预测

摘　　要：目的研究风心病合并房颤患者心肌microRNAs的表达差异,预测其靶基因并分析其可能的生物学功能。方法经知情同意,整群采集2013年1—12月收治的14例住院风心病房颤患者和8例风心病无房颤患者右心耳组织;提取总RNA进行miRNAs芯片杂交检测,应用RMA和FC法筛选差异表达的miRNAs,并用RT-PCR进行验证;运用生物信息学软件预测靶基因并分析其生物学功能。结果与风心病无房颤组相比,风心病合并房颤组有22个miRNAs表达有差异,其中6个miRNAs表达上调,16个miRNAs表达下调;验证结果表明与风心病无房颤组比较,miR-661、miR-520d-5p和miR-145-5p表达显著下调(P<0.01);经GO和KEGG数据分析,差异miRNA靶基因的一部分亦与心肌纤维化相关。结论该研究获得与房颤相关的差异miRNAs,可能是房颤新的生物标志物和潜在治疗靶点。Objective To research the expression difference of myocardium micro RNA of patients with rheumatic and atrial fibrillation and predict the target gene and analyze the potential biology function. Methods Auricula dextra tissues of 14 cases of inpatients with rheumatic and atrial fibrillation and 8 cases of patients were group selected with rheumatic but without atrial fibrillation from January to December 2013 were collected, and the total RNA was extracted for mi RNAs hybridization test, and the mi RNAs was screened by RMA and FC, and then RT-PCR was used for verification, and the target gene was predicted by the biology information software and the biology function was analyzed. Results The expression of 22 mi RNAs had differences in the combined group, including 6 whose expression increased and 16 whose expression decreased, and the expressions of mi R-661, mi R-520d-5p and mi R-145-5p obviously decreased compared with the other group, all P〈0.01, the GO and KEGG data analysis showed that the some of differential mi RNA target genes was correlated with the myocardial fibrosis. Conclusion The differential mi RNA related to atrial fibrillation may the new biomarkers of atrial fibrillation and potential treatment target point.
