摘  要:目的观察Morris水迷宫训练对老年痴呆(AD)大鼠学习记忆能力及海马组织肿瘤坏死因子(TNF)-α、超氧化物歧化酶(SOD)、丙二醛(MDA)的影响。方法随机将32只SD雄性大鼠分为假手术组、模型组、Morris水迷宫短期训练组和Morris水迷宫长期训练组;以穹隆-海马伞切断建立痴呆大鼠模型;运用Morris水迷宫进行训练;采用跳台实验以及水迷宫实验测试大鼠的学习和记忆能力;采用酶联免疫吸附法(ELISA)测定大鼠海马组织TNF-α的水平,比色法检测大鼠海马组织SOD活性和MDA含量。结果 Morris水迷宫训练后AD大鼠学习记忆能力显著提高(P<0.01);大鼠海马区TNF-α、MDA含量明显降低(P<0.05)、SOD含量明显升高(P<0.05)。结论 Morris水迷宫训练能明显改善大鼠的学习记忆能力,其机制可能与提高海马区SOD含量、降低TNF-α和MDA水平有关,而且水迷宫训练时间越长,恢复越明显。Objective To investigate the protective effect of water maze training on Alzheimer,s disease(AD)rats.Methods 32 SD rats were randomly divided into sham,model,short-term Morris water-maze training( short-term) and long-term Morris water-maze training( long-term) groups.Rat model of AD was made by transection of rat fimbria-fornix. Rats in these groups were tested for spatial learning and memory ability in Step-down Test and Morris water maze.Then,the brains were removed and hippocampal homogenate were prepared.Enzyme linked immunosorbent assay( ELISA) was adopted to measure the level of TNF-α,colorimetric method was used to detect the content of superoxide dismutase( SOD) and malondialdehyde( MDA) in the hippocampus homogenate. Results Morris water-maze training,learning and memory ability of AD rat was increased obviously,TNF-α and MDA were reduced,SOD was increased in hippocampal homogenate( P〈0. 05).Conclusions Water maze training can improve the spatial learning and memory ability of AD rats made by transaction of fimbria-fornix by enhancing the content of SOD and reducing TNF-α and MDA levels,and longer training and better effect.
