摘 要:为了探究不同纤维素分解菌的酶活力,获得分解能力更优菌株,试验对从蘑菇渣中分离到的20株能分解纤维素的菌株分别进行了纤维素降解圈的测定、羧甲基纤维素酶活性的测定、滤纸条降解试验以及四种酶(全酶、内切酶、葡萄糖苷酶及外切酶)活力的测定。结果表明:纤维素降解圈试验中,20株菌株中除3个菌株未形成菌落或透明圈外,其他菌株透明圈与菌落直径比值(D/d)均在2.26~5.72之间;18株菌株的羧甲基纤维素酶活力为8.357~330.860 U/g,但不同菌株酶活力差异很大,F1-2、F1-7、F2-4、ZH-3、ZH-4、ZH-5及ZH-6菌株酶活力在100 U/g以上;F1-2、F1-3、F2-3、F2-4、F2-5、ZH-2、ZH-4、ZH-5、ZH-6滤纸条降解阳性。结合全酶、内切酶、葡萄糖苷酶及外切酶活力检测结果综合评判,筛选出F1-2、F1-7、F2-4、ZH-3、ZH-4、ZH-5、ZH-6这7株对天然纤维素有较强降解能力的菌株。In this study, the cellulase activities of cellulose - degrading bacteria isolated from mushroom residue are detected and analyzed. The results indicated that the ratio of transparent circle and colony diameter of other strains were between 2.26 and 5.72, except for three ;strains which did not form colonies or cellulose - solventzonas in the medium containing Congo red; the values of carboxymethyl cellulose activity of eighteen strains were between 8. 357 and 330. 860 U/g; the values of earbexymethyl cellulose activity exhibited great difference among the various strains; the values of carboxymethyl cellulose activity of seven strains ( F1 - 2, F1 - 7, F2 - 4, ZH - 3, ZH - 4, ZH-5 and ZH - 6) were over 100 U/g, while nine strains (F1 -2, F1 -3, F2 -3, F2 -4, F2 -5, ZH-2, ZH-4, ZH-5 and ZH -6) showed positive results in filter paper test; seven strains ( F1 -2, F1 -7, F2 -4, ZH -3, ZH -4, ZH -5 and ZH -6) with high cellulose - degrading effieieneies were obtained based on the results of Congo red cellulose hydrolytic hole measurement, filter paper degradation test and cellulose activity assays( filter paper enzyme FPA, endoglucanase CMCase, exoglucanase C1 and glucosidase 13 - Gase) by using CMC - Na as sole carbon source.
