摘 要:为了提高不动杆菌对石油烃的降解能力,采用离子诱变育种技术,使用生物改性离子注入设备对不动杆菌诱变育种.通过研究不动杆菌a8在不同剂量N+离子注入后存活率的变化趋势,结合气相色谱法(GC)测定得到的N+离子注入前后不动杆菌对石油烃的降解率,获得了离子注入不动杆菌诱变育种的最佳工艺参数.研究结果表明:N^+离子注入剂量与不动杆菌存活率之间呈现出较明显的"马鞍形"变化趋势;离子注入后不动杆菌对石油烃的降解率均有所提高,降解率最高可达95.03%;N^+离子的注入能量和注入剂量分别为15keV和8.0×10^(15) ions·cm^(-2)是不动杆菌离子诱变的最佳工艺参数.To improve the ability of petroleum hydrocarbon-degrading, the acinetobacter was induced mutation by the biological modified ion implantation equipment. The variation of the survival rate of acinetobacter a8 after different doses of N^+ ion implantation was researched,and the degradation rates of petroleum hydrocarbon was measured by gas chromatograph (GC). The optimum implantation parameters of the acinetobacter a8 were determined. The results showed that there was a significant " saddle-shaped" trend between N^6 ion implantation dose and acinetobacter survival rate. The degradation rate of petroleum hydrocarbon increased as high as 95.03% after ion implantation. The injection energy and implantation dose of N+ ion were 15 keV and 8.0×10^15 ions·cm^-2,respectively,which are the optimum process parameters of acinetobacter ionization mutagenesis.
