摘 要:为筛选林芝松口蘑(Tricholoma matsutake)子实体中具抗肿瘤作用的组分,采用MTT法测定林芝松口蘑乙醇提取物的不同有机溶剂萃取分部对乳腺癌细胞(MCF-7)、胃癌细胞(SGC-7901)和肺癌细胞(A549)的体外抑制率。结果表明:林芝松口蘑乙醇提取物的抑癌活性物质主要集中在氯仿分部,100μg/mL作用48h,对MCF-7和SGC-7901的抑制率分别达到了64%和55%;乙醇提取物的各萃取分部对A549抑制效果较差,在实验浓度范围内抑制率均未超过25%。Tricholoma matsutake fruit bodies from Linzhi, Tibet (2 kg) were suspended in 15 volumes of 95% ethanol and extracted under reflux at 85 ℃ for 2 h. After filtration, the residue was twice more subjected to the same procedure and the combined extracts were evaporated to dryness. An aliquot (20 g) of this ethanolic extract was then sequentially extracted with 100 mL of petroleum ether, chloroform, ethyl acetate, nbutanol and double-distilled water, and the extracts evaporated to dryness in each case. Extracts were then tested for their capacity to inhibit the proliferation of the cancer cell lines A549 (lung), MCF-7 (breast) and SGC-7901 (gastric) using the MTT assay. Anti-tumor components present in the T. matsutake fruit bodies were located mainly in chloroform extracts, with MCF-7 and SGC-7901 inhibition rates of 64% and 55%, respectively recorded after 48 h exposure. Proliferation of A549 cells was also inhibited by all the extracts although inhibition rates never exceeded 25% over the range of concentrations adopted.
