VEGF抑制剂阿西替尼对人胃癌细胞周期及凋亡的影响

摘　　要：目的研究VEGF抑制剂阿西替尼(Axitinib)对人胃癌细胞AGS细胞周期和凋亡的影响。方法采用流式细胞仪检测0μmol/L、0.1μmol/L、1μmol/L、5μmol/L的阿西替尼作用AGS细胞24 h后对细胞周期的影响;采用同样方法检测该药对AGS细胞凋亡的影响。结果阿西替尼将AGS细胞抑制在G2/M期,并呈浓度依赖性,差异有统计学意义(P<0.05)。阿西替尼引起AGS细胞的凋亡,随着给药浓度增加,AGS细胞凋亡率也随之增加,差异有统计学意义(P<0.05)。结论阿西替尼通过把胃癌AGS细胞阻滞在G2/M期而抑制细胞的增殖,并促进胃癌AGS细胞凋亡。Objective To study the effect of VEGF inhibitor - Axitinib on the cell cycle and apoptosis of the AGS cell. Method The effect of axitinib on the cell cycle and apoptosis were analyzed using the flow cytometer by double staining with annexinV and propidium iodide. Results Axitinib can induce the cell - cycle of the AGS cell in G2/M phase and can induce the apoptosis in AGS cell, which were concentration - dependent. Conclusion Axitinib can inhibit the proliferation of AGS by arresting the cells in G2/M phase and inducing the apoptosis.
