摘 要:目的探讨干扰c-myc基因表达对人胰腺癌细胞SW1990对化疗药物敏感性的影响。方法采用实时定量PCR和Western blot检测人胰腺癌细胞系SW1990中c-myc基因的干扰效果;MTT法检测癌细胞对化疗药物吉西他滨和卡铂的敏感性;流式细胞术检测吉西他滨诱导的细胞凋亡情况。结果 c-myc si RNA显著下调人胰腺癌SW1990细胞中c-myc基因的m RNA和蛋白表达水平。干扰c-myc表达后,化疗药物吉西他滨和卡铂对SW1990细胞的半数抑制浓度(IC50)均显著减小(P<0.05),吉西他滨诱导的SW1990细胞凋亡率显著升高(P<0.05)。结论干扰c-myc表达可增强人胰腺癌细胞株SW1990对化疗药物吉西他滨和卡铂的敏感性。Objective To investigate the effects of c-myc gene interference on the chemotherapeutic drugs sensitivity in human pancre- atic cancer cell line SW1990. Methods Quantitative real-time PCR and Western blot were used to verify the c-myc gene interference in human pancreatic cancer cell line (SW1990) after small interfering RNA (siRNA) transfection. MTT assay was used to detect the sensitivity changes of the cancer cells to gemcitabine and carboplatin, and flow cytometry was used to evaluate the cell apoptosis induced by gemcitab- ine. Results Results of PCR and Western blot showed that the expression levels of c-myc mRNA and protein were significantly decreased in SW1990 cells transfected by small interfering c-myc (c-myc siRNA). After c-myc gene interference, the median inhibitory concentra- tion of gemcitabine and carboplatin for SW1990 cells were significantly decreased (P〈0.05). The apoptosis rate of SW1990 cells induced by gemcitabine was dramatically increased after c-myc gene interference (P 〈 0.05). Conclusion Inhibition of c-myc gene in human pancreatic cancer cell line SW1990 enhanced its sensitivity to chemotherapeutic drugs gemcitabine and carboplatin.
