摘 要:目的:探讨铁结合蛋白对镉损伤的骨髓间充质干细胞(MSCs)向成骨细胞分化的保护作用。方法:试验被分成对照组、模型组和蛋白干预组。分组处理后,经成骨诱导培养,透射电镜观察细胞外基质形成及超微结构,茜素红染色观察钙结节形成能力,定量PCR法检测波形蛋白(Vimentin)、Run转录因子2(Runx2)、I型胶原蛋白(Col-1)基因m RNA表达水平。结果:MSCs经铁结合蛋白保护,成骨诱导培养21d后进行茜素红染色,细胞钙结节大小、颜色均显著性高于模型组。透射电镜观察MSCs超微结构损害明显得到缓解。Vimentin基因的表达水平先升高后降低(P<0.05),而模型组随时间的延长,表达水平显著性减少(P<0.01)。培养3d和6d的Runx2基因,培养6 d和12 d的Col-1基因都显著性高于模型组(P<0.05),与对照组一致。结论:铁结合蛋白能提高镉损伤的保护作用,促进MSCs向成骨细胞分化的能力,其作用可能是通过上调波形蛋白、Run转录因子2、Col-1基因的表达。Objective: To elucidate whether cell differentiation of Mesenchymal Stem Cells (MSCs) against cadmium (Cd) was protected by Ferritin. Methods: MSCs were randomly divided into three groups: the control group, model group and Ferritin group. After treatment with osteogenic culture medium in different process, the ultrastructure were analyzed by transmission electron microscope (TEM), the calcium nodule formation ability was detected by alizarin red staining, and the mRNA expression of Vimentin, Runx2, Col-1 were assayed by using Real-time PCR. Results: After retreated with Ferritin, MSCs differentiated for 21 days. Alizarin red staining could find that the calcium nodule size and color were significantly higher than the model group. TEM showed that the ultrastructure damage of MSCs was significantly alleviated. What's more, the Vimentin gene expression in ferritin group and the control group were increased and then decreased (P〈0.05), while the model group was significantly reduced in a time period (P〈0.01). The Runx2 gene expression in third day and sixth day, and the Col-1 gene expression in sixth and twelfth day were significantly higher than the model one, so was the control one. Conclusion: Ferritin could improve the protective effect of Cd damage and promote the differentiation of MSCs, which might be connected with the regulation expression gene of Vimentin, Runx2, Col-1.
