摘 要:目的探讨茶多酚对人前列腺癌PC-3M细胞侵袭力的影响。方法在人前列腺癌PC-3M细胞培养器皿中分别加入用完全培养液稀释的40、60、80μg/ml茶多酚溶液,24、48h后通过细胞侵袭实验观察PC-3M细胞的侵袭能力,采用RT-PCR和Western blot检测其MMP-2、TIMP-2 mRNA、蛋白表达水平,并与只添加完全培养液的0μg/ml组比较。结果茶多酚处理后的PC-3M细胞侵透基质胶,迁移至胶中或胶下表面的数量减少,且呈浓度、时间依赖性。与0μg/ml组相比,40、60、80μg/ml组MMP-2mRNA、蛋白表达水平均明显下调,TIMP-2 mRNA、蛋白表达水平均明显上调;茶多酚可在mRNA、蛋白水平下调MMP-2、上调TIMP-2基因转录,大致呈浓度、时间依赖性(均P<0.05)。结论茶多酚可在体外减弱人前列腺癌PC-3M细胞的侵袭力,可能与下调MMP-2、上调TIMP-2基因的转录与表达有关。Objective To investigate the effect of tea polyphenols(TPs) on invasive ability of human prostate cancer PC-3M cells.Methods Human prostate cancer PC-3M cells were treated with 40, 60, 80μg/ml tea polyphenol for 24 h or 48 h in vitro. The invasive ability of PC-3M cells was determined by Transwell assay, and the expression of MMP-2, TIMP-2 mRNA and protein was detected by RT-PCR and Western blot, respectively. Results Transwell assay showed that the invasive ability of PC-3M cells was decreased after treatment with tea polyphenol. The expression of MMP-2 mRNA and protein in PC-3M cells was increased after treated with 60μg/ml and 80μg/ml for 24 h or48h were significantly decreased(P〈0.05), while the expression of TIMP-2 mRNA and protein was increased(P〈0.05) in a dose-dependent and time-dependent manner(P〈0.05). Conclusion Tea polyphenols can reduce the invasive ability of human prostate cancer PC-3M cells in vitro, which may be related to the down-regulation of MMP-2 and up-regulation of TIMP-2 gene expression.
