摘 要:目的观察番茄红素预处理对肢体缺血再灌注后肠黏膜的保护作用,并探讨其可能的作用机制。方法健康成年SD大鼠24只,雌雄不限,随机分为假手术组(A组)、模型组(B组)、番茄红素低剂量组(C组)、番茄红素高剂量组(D组)。C组灌胃番茄红素5mg/kg,D组灌胃番茄红素25mg/kg,A组和B组灌胃等量色拉油,连续7d。采用止血带捆绑大鼠左后肢,建立缺血3h后,再灌注18h肢体缺血再灌注损伤大鼠模型,A组大鼠以同样的方法进行造模,但不予结扎。打开腹腔,取回肠末端肠段固定,并于腹主动脉取血,离心保存。HE染色观察各组大鼠肠黏膜的形态变化,测定血清二胺氧化酶(DAO)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果与A组比较,B组大鼠肠黏膜损伤明显加重,血清DAO和MDA含量明显增加(P<0.01),SOD活性降低(P<0.05);与B组比较,C组大鼠肠黏膜损伤减轻(P<0.01),血清DAO和MDA含量降低(P<0.05);D组大鼠肠黏膜损伤减轻,DAO含量减少(P<0.01),血清SOD活性升高,血清MDA含量减少(P<0.05)。与C组比较,D组血清DAO含量减少,血清MDA含量降低(P<0.05);血清SOD活性升高,肠黏膜损伤稍减轻,但差异无统计学意义(P>0.05)。结论番茄红素可通过抗氧化减轻肢体缺血再灌注后肠黏膜的损伤程度。Objective To observe the protective effects of lycopene pretreatment on the intestinal mucosa after limb ischemia reperfusion injury and explore its mechanism.Methods 24 healthy adult Sprague-Dawley rats were randomly divided into four groups including sham operation group(group A),model group(group B),low-dose lycopene group(group C)and high-dose lycopene group(group D),and each group consists of 6rats.Groups C and D were pretreated with lycopene each day with the dose of 5and 25mg/kg respectively for 7days in succession,while the other two groups were administrated with the equal volume of salad oil respectively.The left hind limbs of all the rats were tied with the tourniquet to induce LI/R injury in rats after 3hours of ischemia and 18 hours of reperfusion,but the rat models of Grouop A were established without ligation.The terminal ileum was obtained to evaluate histopathological changes of intestinal mucosa by HE staining,and the blood was reserved after centrifugation and assayed to determine the activity of serum diamine oxidase(DAO)and superoxide dismutase(SOD)and the level of malondialdehyde(MDA).Results Compared with the rats in group A,the mucosal injury of the rats in Group B was significantly aggravated in light microscopic evaluations,the levels of DAO and MDA were significantly increased(P〈0.01),and the activity of serum SOD was significantly decreased(P0.05)after LI/R.In comparison with the rats in Group B,the mucosal injury of the rats in Group C reduced significantly(P0.01),the levels of DAO and MDA decreased significantly,and the activity of SOD increased significantly(P0.05);the mucosal injury of the rats in Group D reduced,the levels of DAO decreased significantly(P〈0.01),the activity of serum SOD enhanced,and the levels of MDA decreased significantly(P〈0.05).Compared with the rats in group C,the serum DAO levels of the rats in Group D decreased and the activity of MDA decreased significantly(P〈0.05),while the activity of serum
