摘 要:实验设计了一个高效液相色谱法分析咯菌腈杀菌剂的教学实验,通过优化波长、流速、流动相等色谱条件和定性定量分析,使学生更好地理解和掌握高效液相色谱分析技术,通过实例检测和模拟咯菌腈杀菌剂处理樱桃的实验,发挥学生的主观能动性,提高分析问题和解决问题的能力。实验以C18(4.6mm×250mm,5μm)为分离色谱柱,乙腈-水(75∶25)为流动相,流速为1 mL/min;检测波长为270nm。该方法分析时间短,峰形较好,且目标物与样品杂质完全分离,适合樱桃中咯菌腈的检测。Teaching experiment was designed to detect fludioxonil by high performance liquid chromatography( HPLC). Through the experiment including wavelength,flow rate,mobile phase,qualitative and quantitative analysis,students could understand and master HPLC. By way of example testing and simulation,play the initiative of students,improve the ability to analyze problems and problem-solving. Reversed phase column used C18( 4. 6 mm×150 mm,5 μm),mobile phase were consist of water( 25%) and acetonitrile( 75%). The flow rate was 1 mL/min,detection wavelength set at 270 nm.This method is short time,better peak shape,complete separation,and can be applied to fludioxonil detection in cherries in vegetables.
