摘 要:蜗牛酶存在于蜗牛的嗦囊和肠道中,是由微生物产生的混合酶,用来帮助宿主消化食物中的多聚物。蜗牛酶广泛应用于细胞生物学和基因工程学实验及饲料添加剂上,现只能从蜗牛体内提取,开发微生物发酵法生产蜗牛酶很有必要。该研究用平皿筛选法从腹足纲动物灰巴蜗牛的粪便中筛选能降解植物常见多聚物的微生物,结果分离到一株放线菌——GM。初步研究发现,它具有分泌甘露聚糖酶、蛋白酶、葡聚糖酶、几丁质酶、果胶酶、阿拉伯树胶酶、木聚糖酶、淀粉酶、纤维素酶等多种酶的能力,并从形态、生理、生化、16SrDNA序列等方面对分离菌进行分析,已明确其分类地位,归属于放线菌科的纤维化纤维菌,相似度在100%。Snailase is a multi-enzyme produced by microorganisms and existing in the crop and intestines of a snail to assist their hostdigest polymers in the food. Snailase, which is widely used in cytobiology and genetic engineering experiments and feed additives, canonly be extracted from snails at present. It is necessary to develop a microbiological fermentation method for producing snailase. In thisstudy the plate external screening method was adopted to screen out microorganisms capable of degrading common plant polymers. Astrain of actinomyces named GM was isolated which the preliminary study found out is capable of secreting many enzymes includingmannase, protease, glucanase, chitinase, pectinase, gum arabic polysaccharide enzyme, xylanase, amylase and cellulase. An analysis wasmade on the isolated strain from such aspects as morphology, physiology, biochemistry, 16S rDNA sequence and the taxonomic status ofthe strain was clearly defined as Cellulosimicrobium cellulans in Actinomycetaceae with 100% similarity.
