摘 要:【目的】研究沙丁胺醇在肉牛血浆、尿液中残留消除规律.【方法】选取3头中国西门塔尔杂交肉牛,连续饲喂沙丁胺醇21d,给药剂量0.45mg/(kg·d),使用UPLC-MS/MS法分别测定给药第1、7、14、21天和停药第3、7、14、28天酶解前、后的血浆和尿液样品中沙丁胺醇质量浓度.【结果】给药期内,酶解前、后血浆中沙丁胺醇质量浓度范围分别为0.23~0.32ng/mL、34.2~42.48ng/mL,停药后迅速消除,分别在停药第3天(未酶解血浆)和停药第14天(酶解后血浆)时低于LOQ(0.2ng/mL),而且酶解后血浆中沙丁胺醇质量浓度显著高于酶解前(P<0.01);给药期内,酶解前、后肉牛尿液中沙丁胺醇含量分别在给药第21天(2 444.4ng/mL)和给药第14天(7 863.4ng/mL)达到峰值,随后逐渐消除,停药28d时尿液中沙丁胺醇浓度低于LOQ(0.5ng/mL),而且酶解后尿液中沙丁胺醇质量浓度显著高于酶解前(P<0.01).【结论】与血浆相比,尿液中沙丁胺醇质量浓度较高,消除速率较慢,而且酶解样品中的沙丁胺醇质量浓度显著高于未酶解样品.【Objective】The aim of this study was to evaluate the concentration of salbutamol residues in plasma and urine samples of cattle during and after treatment.【Method】Three Chinese Simmental cattle continuously received an oral administration of 0.45mg/(kg·d)salbutamol for 21 days.Salbutamol concentrations(before and after hydrolysis)were determined at1,7,14,and 21 dof administration and at 3,7,14,and 28 dafter drug withdrawal using UPLC-MS/MS.【Result】During treatment period,salbutamol concentrations were 0.23-0.32ng/mL and 34.2-42.48ng/mL in plasma samples before and after hydroly-sis,respectively,then decreased rapidly on withdrawal time,and were below the limit of quantitation(0.2ng/mL)at 3and 14 dafter withdrawal,respectively.The concentrations of salbutamol in plasma samples before hydrolysis was significantly higher(P〈0.001)than that in plasma samples after hydrolysis.The peaks of salbutamol urine concentration appeared on treatment days of 21(unhydrolyzed samples2 444.4ng/mL)and 14(hydrolyzed samples 7 863.4ng/mL),respectively,then eliminated gradually,and was undetectable at 28 dof withdrawal.The concentrations of salbutamol in hydrolyzed urine samples were significantly higher(P〈0.01)than those in unhydrolyzed urine samples.【Conclusion】Compared with plasma,salbutamol concentrations in urine samples was higher and eliminated slowly.The concentrations of salbutamol in hydrolyzed samples was significantly higher than that in unhydrolyzed samples(P〈0.01).
