摘 要:目的:优化玛咖总黄酮树脂纯化工艺参数。方法:以玛咖总黄酮纯度为指标,通过静态——动态吸附试验,确定树脂纯化工艺的最佳参数。结果:AB-8大孔树脂分离纯化黄酮的最佳工艺参数为:以质量浓度为0.84g/m L的药液进行上柱,上样液p H为:3~4,洗脱流速为:1m L/min,除杂溶剂为:以蒸馏水冲至Molish反应呈阴性和4BV的20%乙醇,洗脱溶剂为:收集70%乙醇洗脱液2~4BV,总黄酮纯度可达15.73%。结论:工艺稳定、简单、重现性好,可用于玛咖总黄酮的富集。Objective : To investigate the purification technology of total flavonoids from Maca by macroporous resin. Methods : The optimum process parameters of resin were chosen with static and dynamic adsorption and desorption experiments and used the purity of flavonoids as the index. Results: The optimal technology parameters of AB-8 resin were as follows, pH 3 to 4, flow velocity of loading sample lmL/min, distillated water washing till Molish reaction of negative, 4BV 20% ethanol, 70% ethanol elution 2 - 4BV. Under the above conditions, the purity of total flavonoids was up to 15.73%. Conclusion : This purification technology was stable, simple and with good repeatability. It could effectively enrich Maca total flavonoids.
