摘 要:采用40%丙酮提取灰树花子实体中的多酚类物质,经NKA-Ⅱ型大孔吸附树脂纯化,然后依次用乙酸乙酯、正丁醇萃取制备不同极性的多酚组分。通过分析总还原力、DPPH自由基清除率、羟自由基清除率、亚铁离子螯合能力,从而研究不同极性多酚组分的抗氧化活性。结果表明,不同多酚质量浓度的乙酸乙酯相总还原力均高于正丁醇相和水相;乙酸乙酯相的DPPH自由基清除率IC50值为12.192μg/m L,且羟自由基清除率IC50值为66.040μg/m L,说明乙酸乙酯萃取相中所含的多酚组分抗氧化活性较强。Polyphenols of Grifola frondosa fruiting body are extracted by 40% acetone in this study. The extracted polyphenols are purified by NKA-II macroporous resin. Different polaritied polyphenols components are extracted with ethyl acetate and n-butanol successively. Using reducing power, DPPH free radical scavenging rate, hydroxyl free radical scavenging rate and ferrous ion chelating power (FICP) as indexes, the antioxidant activity of the different polarities components are determined. The results show that reducing power of ethyl acetate phase is higher than n-butanol phase and aqueous phase, and IC5 0 val-ues of DPPH free radical scavenging rate and hydroxyl free radical scavenging rate at ethyl acetate phase are 12.192 pg/mL and 66.040 pg/mL respectively, revealing antioxidant activities of polyphenol components extracted with ethyl acetate are stronger.
