5-羟色胺受体相关基因的交互作用与抑郁症自杀行为的关联研究

摘　　要：目的探讨抑郁症患者自杀行为与5-HT1BR基因（rs6298），5-HT2AR基因（rs6311、rs6313）及5-HT2BR基因（rs765458）交互作用的关联。方法提取281例伴随冲动性自杀行为的抑郁症患者（病例组）和281名身心健康的对照者（对照组）基因组DNA，采取TapMan探针单核苷酸多态性（SNP）基因分型的技术检测病例组及对照组所选取的位点基因型；采用x^2检验分析病例组和对照组的基因型和等位基因的分布差异；采用Haploview4．0软件实施单倍型和连锁不平衡分析，GM—DR分析基因．基因交互效应与自杀行为的关系。结果5-HT2AR基因的rs6313和rs6311位点存在连锁不平衡（D-0．756，r^2=0．375），rs6313、rs6311存在单体型。5-HT1BR基因rs6298，5-HT2AR基因rs6311、rs6313及5-HT2BR基因rs765458交互作用，检验准确度为O．6182，CV值为10／10，差异有统计学意义（P〈0．05）。结论5-HT2AR基因的位点rs6311、rs6313单体型与自杀行为可能存在关联。5-HT1BR基因rs6298，5-HT2AR基因rs6311、rs6313及5-HT2BR基因rs765458交互作用与抑郁症自杀行为可能存在某些关联。Objective To investigate the single nucleotide polymorphisms （SNPs） of 5-HT1BR （ rs6298 ）, 5-HT2AR （ rs6311, rs6313 ） and 5-HT2BR （ rs765458 ） gene, and their gene-gene interactions on suicidal behavior in major depressive disorder. Methods The blood samples were taken from 281 depression patients with impulsive suicide attempt and 281 age-matched healthy controls from a hospital in Harbin city, Heilongjiang province.The DNA isolated from blood samples and was genotyped using TapMan SNP genoty- ping probe.X2 test was used to compare differences in the distribution of gene alleles between cases and con- trols.Haplotype and linkage disequilibrium（LD） analysis was performed using Haploview 4.0 software.GMDR was used to analyze the gene-gene interaction. Results The rs6313 and rs6311 of the 5-HT2AR gene were in strong linkage disequilibrium （ D = 0.756, r^2 = 0.375 ）. There was a significant gene-gene interaction of 5-HT 1BR （ rs6298 ）, 5-HT2AR （ rs6311, rs6313 ） and 5-HT2BR （ rs765458 ） on suicidal behavior （P〈 0.05 ）. In this model,the test accuracy was 0.6182 and CV value was 10/10.Conclusion A haplotype containing rs6311 and rs6313 of 5-HT2AR gene is associated with suicidal behavior.The interaction of 5-HT1BR gene （rs6298） , 5-HT2AR gene （ rs6311, rs6313） and 5-HT2BR gene （rs765458） are associated with depression suicidal behavior.
