摘  要:以棉籽饼粉为试验材料,花生、大豆和菜籽饼粉为对照,在分析其化学组分的基础上,利用超声波辅助碱法提取棉籽蛋白,研究超声波处理参数对棉籽蛋白提取率的影响。在单因素试验基础上,设计Box-Behnken试验优化超声波辅助碱法提取棉籽蛋白的工艺条件。结果表明,棉籽饼粉蛋白含量高(61.89%),可作为植物蛋白资源。棉籽蛋白提取优化工艺参数为pH 11.0、液料比20∶1 m L/g、超声时间20 min、超声温度45℃、超声波功率600 W,蛋白提取率可达87.64%。所建棉籽蛋白提取回归模型高度显著(R2=0.995 6),拟合性好,可用于预测棉籽蛋白的提取率。与常规碱性法相比,超声波辅助碱法提取棉籽蛋白所需时间短,温度低,提取率高。Compared with peanut, soybean and rapeseed meal, contents of chemical components of cottonseed meal were analyzed, and in order to study the effect of ultrasonic treatment parameters on the extraction yield of protein from cottonseed meal, cottonseed protein was extracted by ultrasonic-assisted alkaline extraction method. Based on single factor experiments, the Box-Behnken test was designed to optimize the process conditions of cottonseed protein by ultrasonic-assisted alkaline extraction. The results showed that the content of protein of cottonseed meal was as high as 61.89%, which could be used as protein resource of plants. And the optimum conditions were 45℃ of ultrasonic temperature, 600 W of ultrasonic power, 20 : 1 mL/g of liquid-solid ratio, 11.0 of pH, and 20 min of ultrasonic time. Under the optimum conditions, the extraction yield of cottonseed protein was 87.64%. It proved that the model was highly significant (R2=0.995 6) and it could fit experi- mental data well and be applied in the prediction of extraction yield of cottonseed protein. Compared with the alkaline method without ultrasonic treatment, ultrasonic-assisted alkali extraction method of cottonseed protein showed shorter time, lower temperature and higher extraction yield.
