摘  要:试验比较不同来源白术提取物对水杨酸、间羟基苯甲酸、对羟基苯甲酸、对香豆酸、阿魏酸、丁香酸、肉桂酸、芥子酸和绿原酸9种酚酸物质的高效液相色谱法检测及白术来源对其酚酸物质含量的影响。试验采用ACQUITY UPLC?BEH C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相A相为0.3%乙酸水溶液、流动相B相为90%甲醇水溶液,梯度洗脱,流速为1.0 m L/min,DAD检测波长为285 nm,柱温为40℃。检测方法显示:9种酚酸物质的检测线性范围为0.02~500 mg/L;检出限(S/N=3)在0.002~0.01 mg/kg之间;在加标浓度为10~80 mg/kg条件下,加标回收率(N=9)在77.78%~100.47%之间,检测方法灵敏有效;标准品进样6次的结果RSD均小于5%,表明精密度良好。检测结果表明,不同来源白术提取物得到的白术提取物中水杨酸、间羟基苯甲酸、对羟基苯甲酸、对香豆酸、阿魏酸、丁香酸、肉桂酸、芥子酸和绿原酸9种白术提取物酚酸物质的总含量以浙江新昌产白术提取物最高。In this study, the influence of different Atractylodes macrocephala Koidz extracts on contents of nine kinds of the phenolic acids (salicylic acid, m-hydroxybenzoic acid and p-hydroxybenzoic acid, p-coumaric acid, ferulic acid, cinnamic acid, cinnamic acid, sinapic acid and chlorogenic acid) in Atractylodes macrocephala Koidz extracts was compared by using HPLC. The experiment was based on ACQUITY UPLCBEH C18 column (250 mm ×4.6 mm, 5 μm) with mobile phase A of 0.3% acetic acid aqueous solutionand mobile phase B of 90% methanol aqueous solution, and gradient eluted with the flow rate of 1.0 mL/min, DAD detection wavelength of 285 nm and column temperature of 40℃. Detection method showed sensitive and effective: ninekinds of phenolic acidslinear ranged from 0.02-500 mg/L; The detection limit (S/N=3) was between 0.002-0.01 mg/kg, the recoveries (N=9) was between 77.78%-100.47% in the added concentration range of 10-80 mg/kg. It was shown by the results that the total content of ninephenolic acids (salicylic acid, m-hydroxybenzoic acid, p-hydro^ic acid, p-cournaric acid, ferulic acid, cinnamic acid, cinnamic acid, sinapic acid and chlomgenic acid) in extracts was the highest in Zhejiang Xinchang Atractylodes macrocephala Koidz extracts.
