戊型肝炎疫苗评价用小鼠品系的比较

摘　　要：目的比较不同品系及相同品系不同来源BALB/c小鼠对戊型肝炎疫苗(HE疫苗)的免疫应答,探讨适用于HE疫苗评价的小鼠品系,规范疫苗效力评价用动物。方法分别用2家企业制备的HE疫苗免疫BALB/c、C57BL/6、KM和NIH 4种品系小鼠及3种不同公司来源的BALB/c小鼠,均免疫1针,28 d后采血,分离血清,检测抗体应答,计算效力ED_(50)。结果 2家疫苗免疫C57BL/6、KM和NIH小鼠时,各剂量组均表现出敏感应答,提示C57BL/6、KM和NIH小鼠对本研究中的2家疫苗不能灵敏地反映出剂量变化趋势,不适用于HE疫苗效力评价;BALB/c小鼠对2家企业各剂量组HE疫苗均呈现良好的免疫应答,且呈明显的剂量效应关系。3种不同来源的BALB/c小鼠免疫HE疫苗后,抗体应答存在差异。结论 BALB/c小鼠可作为评价HE疫苗免疫原性和效力ED_(50)的模型动物;应用BALB/c小鼠进行疫苗评价时需固定小鼠来源。Objective To compare the immune responses of BALB/c mice of various strains and from various sources to hepatitis E（HE）vaccine, investigate the mouse strains suitable for evaluation of HE vaccine and normalize the laboratory animals for evaluation of vaccine potency. Methods BALB/c, C57BL/6, KM and NIH mice as well as BALB/c mice from three corporations were immunized once with HE vaccines from two manufacturers separately, of which the serum samples were collected 28 d after immunization and determined for antibody response, and the ED（50） was calculated.Results C57BL/6, KM and NIH mice showed sensitive responses to HE vaccines from two manufacturers at any dosages,indicating that the three mouse strains could not reflect the dosage change tendency thus was unsuitable for evaluation of HE vaccine potency. However, BALB/c mice showed good immune responses to the HE vaccine from two manufacturers at various dosages, and significant dose-response relationship was observed. The antibody responses of BALB/c mice from three corporations to HE vaccine showed difference. Conclusion BALB/c mice may be used as the model animals for evaluation of immunogenicity and potency ED（50） of HE vaccines. However, the source of BALB/c used for vaccine evaluation should be immobilized.
