结直肠癌患者NQO1蛋白检测的临床意义及疾病预后价值分析

摘　　要：目的:本研究旨在检测结直肠癌患者肿瘤组织中NQO1蛋白表达水平及其对患者疾病预后价值分析。方法:采用实时定量PCR(Q-PCR)方法检测NQO1mRNA表达情况,采用Western blotting和免疫组化方法检测NQO1蛋白表达情况,采用siRNA技术干扰结直肠癌细胞系SW480NQO1表达。结果:与正常组相比,siRNA组在24h、48h、72h细胞活力均降低(P<0.05)。与非肿瘤组织相比,结直肠癌组NQO1mRNA和蛋白的表达量均升高(P<0.05)。结直肠癌患者样本为肿瘤组织NQO1阳性率高于临近非肿瘤组织(P<0.05)。NQO1表达与结直肠癌患者肿瘤转移、乙型肝炎病毒(HBV)感染、淋巴结肿瘤转移(TNM)分期及肿瘤直径密切相关(P<0.05)。NQO1阳性结直肠癌患者的总生存期、无疾病生存期均低于阴性患者(P<0.05)。结论:结直肠癌患者肿瘤组织中NQO1蛋白表达上调,且其表达与肿瘤恶性状态密切相关,参与结直肠癌细胞的侵袭和转移,可作为结直肠癌患者疾病预后标志物。Objective：The aim of this study was to evaluate the feasibility of NQO1 as a novel prognostic marker for colorectal neoplasms.Methods：Quantitative real-time PCR was performed to assess mRNA levels of NQO1 in matched colorectal neoplasms specimens.Western blotting assays and immunohistochemistry were performed to assess protein levels of NQO1 in matched colorectal neoplasms specimens.Using siRNA disturbed NQO1 expression in SW480 cells.Results：Compared with the normal control group,cell activities in siRNA group were lower in 24 h,48hand 72 h.NQO1expression was higher in colorectal neoplasms compared to the expression found in adjacent tissues at messenger mRNA and proteins levels（P〈0.05）.Notably,NQO1 expression was significantly higher in colorectal neoplasms compared to adjacent tissues（P〈0.05）.Furthermore,positive NQO1 expression was associated with tumor size,tumor capsulae,status of metastasis,and tumor-node-metastasis stage（P〈0.05）.Compared with the NQO1 negative patients,overall survival（OS）rate and disease-free survival（DFS）in NQO1 positive patients were lower（P〈0.05）.Conclusion：Our results suggest that NQO1 could play an important role in colorectal neoplasms and serve as an independent unfavorable prognostic factor for OS and DFS for colorectal neoplasms.
