摘 要:目的探讨循环IL-27和组织T细胞免疫球蛋白及黏蛋白结构域分子3(Tim-3)在晚期非小细胞肺癌(Non-small Cell Lung Cancer,NSCLC)患者中的表达。方法选取2015年5月—2016年5月在我院就诊的NSCLC患者60例。根据NSCLC分期,分为NSCLC Ⅱ期组、NSCLC Ⅲ期组、NSCLC Ⅳ期组。采用酶联免疫吸附法检测各组研究对象外周血细胞因子IL-27的表达,免疫组织化学法检测非小细胞肺癌组织中Tim-3表达。结果NSCLC患者外周血IL-27表达均存在统计学差异(P<0.05);其中以IL-27浓度在NSCLC Ⅳ期患者中表达最低,在NSCLC Ⅱ期患者中表达最高(P<0.05)。免疫组织化学检测结果分析显示各期NSCLC患者组织Tim-3表达存在统计学差异(P<0.05);其中Tim-3在NSCLC Ⅲ期和Ⅳ期患者表达较NSCLC Ⅱ期患者明显升高(P<0.05)。NSCLC患者分期与外周血IL-27呈负相关(r=-0.723,P<0.01),与组织Tim-3表达呈正相关(r=0.375,P<0.01);外周血IL-27与组织Tim-3表达呈负相关(r=-0.769,P<0.01)。结论晚期NSCLC患者外周血IL-27表达降低、组织中Tim-3表达升高;IL-27和Tim-3是NSCLC的潜在免疫调控靶点。Objective To investigate the expression of circulating IL - 27 and T cell immunoglobulin and mucin - domain - containing molecule - 3 ( Tim - 3) in patients with advanced non - small cell lung cancer ( NSCLC) . Methods 60 patients with NSCLC who were treated in our hospital from May 2015 to May 2016 were enrolled into our study. According to NSCLC stage, all patients were divided into NSCLC stage II group, NSCLC stage III group and NSCLC stage IV group. The expression of IL - 27 in peripheral blood cytokines was detected by enzyme - linked immunosorbent assay (ELISA) and the expression of Tim - 3 in tissues of patients with NSCLC was detected by immunohistochemistry. Results There was signifi-cant difference in the expression of circulating IL -27 among NSCLC patients (P 〈0. 05) . The expression of IL -27 was the lowest in NSCLC stage IV group and highest in NSCLC stage II group (P 〈 0. 05 ). According to the immunohistochemistry results, there was significant difference in the expression of Tim - 3 among three groups ( P 〈 0. 05 ) . The expression of Tim -3 in NSCLC stage III group and NSCLC stage IV group were significantly increased as compared with that in NSCLC stage II group (P 〈0. 05) . The stage of NSCLC was negatively correlated with the IL -27 in peripheral blood (r = -0.123 , P 〈 0. 01) , and positively correlated with the expression of Tim - 3 (r = 0. 375 , P 〈 0. 01) . IL - 27 in peripheral blood was negatively correlated with Tim - 3 expression (r = -0.169 , P〈0. 01). Conclusions The expression of IL - 27 in the pe-ripheral blood decreases and tissue Tim -3 increases in the patients with advanced NSCLC. IL-27 and Tim - 3 are potential immune - regulatory targets of NSCLC.
