摘  要:发菜是一种陆生耐旱固氮蓝藻,具有重要的生态和资源价值,但目前发菜的基因组和转录组信息还比较缺乏。本研究采用Illumina Hi Seq TM 2500高通量测序平台对充分吸水和失水后的发菜藻体进行转录组分析,经过拼接组装共获得30 013 560条clean reads,3 100个Unigene,序列平均长度854 nt。将Unigene序列与NR、NT、Swiss-Port、PFAM、GO、COG、KEGG(E-value<1.0E^(-5))等数据库进行比对,共有2 874个Unigene获得了注释。通过GO功能分类,917个Unigene映射到GO不同功能节点上;通过Unigene与COG数据库的比对,可将有COG功能的514个Unigene分为20类;通过KEGG pathway分析,有1 220个编码基因参与了135条已知的代谢通路。提示谷胱甘肽、类胡萝卜素、淀粉和蔗糖、ABC转运体等代谢途径的部分基因在失水藻体上调表达,说明这些通路与发菜耐旱性关系密切。因此这些注释信息为今后发菜耐旱基因的挖掘提供了重要依据,同时也为发菜分子生物学的进一步研究提供了试验基础。Nostoc flagelliforme is a kind of terrestrial cyanobacterium which shows a strong ecological adaptability to xeric conditions and has important ecological and resources value.However,the genome and transcriptome data of N.flagelliforme is still relatively lacking.In this study,the transcriptomes of rehydrated and dehydrated N.flagelliforme colonies were sequenced by Illumina HiSeqTM2 500 high-throughput sequencing platform.In total30013 560 clean reads and 3 100 Unigenes were obtained through splicing and assembling,with an average sequence length of 854 nt.Compared Unigene sequence with NR,NT,Swiss-Prot,PFAM,GO,COG,and KEGG databases(E-value1.0E^(-5)),2 874 Unigenes were annotated.A total of 917 Unigenes were mapped into different GO terms via Go function classification.Through the comparision between the Unigene and the COG database,the 514 Unigenes which had COG function were divided into 20 classes.KEGG pathways analysis showed that1 220 coding genes were involved in 135 known metabolic pathways.These results indicated that some genes of metabolic pathways such as glutathione,carotenoid,starch and sucrose as well as ABC transporter in the dehydrated colonies were up-regulated,suggesting these pathways might be closely related to drought resistance.Therefore,the annotation information could provide an experimental basis for future identification of drought-tolerant genes of N.flagelliforme,meanwhile it could also lay a foundation for the further study of the molecular biology of N.flagelliforme.
