摘  要:为了比较阪崎克罗诺杆菌在玻璃、聚丙烯及不锈钢表面形成的生物膜,将培养48 h的阪崎克罗诺杆菌生物膜经荧光染料SYTO 9/PI标记后,用激光共聚焦扫描显微镜(confocal laser scanning microscope,CLSM)联合COMSTAT软件定性定量地分析生物膜结构。结果表明:阪崎克罗诺杆菌在3种材料表面形成的生物膜的总生物量、平均厚度及平均扩散距离依次是不锈钢>聚丙烯>玻璃,粗糙系数及表面积与生物量比值则为不锈钢<聚丙烯<玻璃,生物膜的菌体活死比例是玻璃高于聚丙烯、不锈钢。另外,标准菌株ATCC 29544生物膜的总生物量、菌体活死比例、平均厚度及平均扩散距离均高于分离株Ⅰ、Ⅱ,粗糙系数及表面积与生物量比值均低于分离株。可见,阪崎克罗诺杆菌在不锈钢表面的成膜性强于玻璃与聚丙烯,标准菌株ATCC 29544的成膜能力高于2株食品分离菌株。In order to investigate Cronobacter sakazakii.biofilm( 48 h) on the surface of glass slide,polypropylene and stainless steel,the fluorescence image stacks of C.sakazakii biofilm tagged with SYTO 9/PI was assayed with hyphened technique of the program confocal laser scanning microscopy( CLSM) and COMSTAT. The result showed that the sequence from high to low of total biomass,average thickness and average diffusion distance was: stainless steel,polypropylene and glass slide,while the sequence from high to low of roughness coefficient and surface to volume was: glass slide,polypropylene and stainless steel.Live/dead ratio of C.sakazakii biofilm on the surface of glass slide was higher than polypropylene and stainless steel.Compared with strain Ⅰ and Ⅱ,the biofilm of C.sakazakii strain ATCC 29544 had higher level of total biomass,live/dead ratio,average thickness and average diffusion distance,while roughness coefficient and surface to volume had lower level. In conclusion,C.sakazakii produced stronger biofilm on the surface of stainless steel than glass slide and polypropylene,and the biofilm forming ability of strain ATCC 29544 was higher than strain Ⅰ and Ⅱ isolated from food.
