摘 要:目的观察王老吉凉茶的急性经口毒性和30 d经口毒性,为评价其食用安全性提供科学依据。方法急性经口毒性试验:选用18~22 g NIH种小鼠,1次灌胃给予15.0 g/(kg·BW)[折算药材剂量99.9 g/(kg·BW)]王老吉凉茶浸膏,每日观察小鼠的死亡情况,共观察14 d。30 d经口毒性试验:雌雄SD大鼠每日灌胃给予1.88、3.75、7.50 g/(kg·BW)[相当于药材剂量12.5、25.0、50.0 g/(kg·BW)]剂量王老吉凉茶浸膏,对照组灌胃同等容量纯净水,共30 d。实验结束时检测常规生化和血常规指标,称量脏器重量,对照组和7.50 g/(kg·BW)组进行组织病理学检查。结果急性经口毒性试验在观察期内动物无不良反应和死亡,雌雄小鼠急性经口毒性LD_(50)>15.0g/(kg·BW);30 d经口毒性试验中各剂量组受试物对雌雄SD大鼠的体重、增重、摄食量、食物利用率、血常规、血生化、脏器重和脏体比均无明显影响,病理组织学检查也没有发现与王老吉凉茶浸膏相关的改变。结论王老吉凉茶浸膏属无毒级,在7.50 g/(kg·BW)[相当于药材剂量50.0g/(kg·BW)]下未观察到大鼠明显毒性反应。Objective To explore the acute oral toxicity and 30-day toxicity of Wanglaoji herbal tea, so as to provide scientific basis for evaluating its edible safety. Methods Acute oral toxicity test: Wanglaoji herbal tea extract was administered to the NIH mice via oral gavage at doses of 15.0 g/( kg · BW) [ equivalent to herbal crude drugs 99.9 g/( kg · BW) ] ,and symptoms and deaths of NIH mice were observed for 14 days. Thirty-day oral toxicity test: Wanglaoji herbal tea extract was administered to the male and female Sprague Dawley rats via oral gavage at doses of 1.88, 3.75, and 7.50 g,/( kg · BW) [ equivalent to herbal crude drugs 12.5, 25.0, and 50.0 g/( kg · BW) ], and the same volume of purified water was given to rats in control group, for 30 consecutive days. Hematologic and biochemistry tests were performed, and organs were weighed at the end of the experiment. Histopathological examination was con- ducted in the control group and 7.50 g/( kg · BW) group. Results In the acute oral toxicity test, no ad- verse reactions or deaths were observed during the observation period. The LD50 of the acute toxicity was above 15.0 g/(kg · BW). In the 30-day oral toxicity, all dose groups had no obvious effect on body weight, weight gain, food intake, food utilization rate, hematologic tests, biochemical tests, organ weight and the ratios of organs in male and female rats . Histopathological examination did not show any changes associated withWang Laoji herbal tea extract. Conclusion Wanglaoji herbal tea extract belongs to non-tox- ic level, and 7.50 g/( kg · BW) [ equivalent to herbal crude drugs 50.0 g/( kg · BW) ] Wanglaoji herbal tea extract would not cause obvious toxic reaction in rats.
