摘  要:探究大孔树脂吸附法纯化松茸多糖的最佳工艺条件,为食用菌类水溶性多糖的精制提供参考。以松茸多糖得率为指标,通过单因素试验和正交试验,确定大孔树脂纯化松茸多糖的最佳工艺参数。洗脱剂浓度60%,洗脱剂流速5mL/min,上样液浓度0.6mg/mL,上样液pH6,吸附时间1h,解吸时间1h,在此条件下纯化松茸多糖得率达到11.58%。此条件下纯化松茸多糖的纯度为25.1%,较粗提物纯度10.10%提高了2.48倍。该工艺条件科学合理,AB-8大孔树脂的吸附性良好,可有效用于松茸多糖的纯化,提高松茸提取物中的多糖含量。The best purification technology of polysaccharide from Tricholomamatsutake was explored by macroporous resin adsorption method,which provided a reference for refining of water-soluble polysaccharide from edible fungi.Using polysaccharide yield as evaluation index, the optimum parameters were determined by the single factor test and orthogonal test. The purification effect of polysaccharide was best by AB-8 macroporous resin, and the best craft was eluent concentration 60%, eluent flow velocity 5 mL/min, the sample liquid concentration 0.6 mg/mL and pH6, adsorption time 1 h, desorption time 1 h. Under this condition, the polysaccharide yield was 11.58%. In this condition, the purity of the polysaccharide was 25.1%, and the purity of the material was 2.48 times higher.The technology is reasonable. The absorption of AB-8 macroporous resin is good, which can be used in the purification of matsutake polysaccharide and improve the polysaccharide content in matsutake extraction.
