摘 要:【目的】优化施氏假单胞菌液态发酵降解茶皂素的条件,以提高油茶籽粕在饲料方面的应用性能,增加经济效益和社会效益。【方法】以茶皂素降解率为评价指标,利用施氏假单胞菌液态发酵降解茶皂素,首先以培养时间、培养温度、培养基初始pH值、瓶口纱布层数、接种量为单一变量进行单因子试验,然后通过Plackett-burman实验、最陡爬坡试验、Box-Behnken设计响应面试验对施氏假单胞菌液态发酵条件进行优化。【结果】单因素试验获得的最佳单因素分别为:培养时间6d,培养温度25℃,培养基初始pH为8,瓶口纱布层数为6层,接种量为1%。在条件优化以后最佳的液态发酵条件为:培养时间5d,培养温度23℃,培养基初始pH为8,瓶口纱布层数为6层,接种量为1%,在此条件下,施氏假单胞菌液态发酵降解茶皂素的降解率达到66.92%。【结论】施氏假单胞菌微生物发酵法降解茶皂素具有成本低、重复性好、无二次污染等明显优势,在饲料方面具有一定的利用价值。【Objective】This study optimized the conditions to degrade tea saponin by Pseudomonas stutzeri liquid fermentation to improve the application performance of tea seed meal in feed and increase economic and social benefits.【Method】Pseudomonas stutzeri was used to degrade tea saponin using the degradation rate as evaluation index.The optimum fermentation conditions were determined by single factor fermentation experiments including time,fermentation temperature,initial pH of culture medium,the number of layers of gauze on the top of bottle and inoculum size.Then,the fermentation conditions were optimized by the Plackett-burman experiments,Steepest ascent experiment and Box-Behnken design response surface test.【Result】The best single factor conditions were:6 days,25℃,pH=8,6 layers,and 1%inoculum size,respectively.The optimum conditions of fermentation degradation overall were 5 days,23℃,pH=8,6 layers,and 1%inoculum size.Under the optimal conditions,the decomposition rate of tea saponin was 66.92%.【Conclusion】Microorganism fermentation degradation of tea saponin with Pseudomonas stutzeri had the characteristics of low cost and low equipment requirement,good repeatability,no secondary pollution and valuable.
