摘 要:目的探讨不同浓度的辣椒素对食管鳞状细胞癌EC109细胞的体内外研究。方法不同浓度的辣椒素(低剂量组5mg/kg,中剂量组10mg/kg,高剂量组20mg/kg)处理食管癌EC109细胞48h后,CCK-8法检测细胞增殖,流式细胞术检测细胞周期;建立食管癌细胞EC109的裸鼠皮下移植瘤模型,Tunnel法检测细胞凋亡指数,免疫组化法检测各组瘤体内Ki-67的阳性表达指数。结果不同浓度的辣椒素对食管鳞状细胞癌EC109细胞均具有抑制细胞增殖的作用,还可扰乱细胞的正常周期,G0/G1期细胞数目相应增加,并呈现明显的浓度依赖现象;体内实验部分,运用Tunnel法、免疫组化法检测各组瘤体内Ki-67的阳性进一步说明辣椒素对食管鳞状细胞癌EC109细胞具有抑制增殖、促进凋亡的作用。结论不同浓度的辣椒素均具有抑制食管鳞状细胞癌EC109细胞增殖及促进凋亡的作用。并呈现明显的浓度依赖现象。Objective To evaluate the study of the different concentrations of capsaicin on the ECI09 cells of esophageal squamous cell carcinoma in vitro and in vivo. Methods The EC109 cells were treated with the different concentrations of capsaicin (the low dose group of capsaicin was 5mg/kg, the middle dose group of capsaicin was 10mg/kg, the high dose group of capsaicin was 20mg/kg. ) for 48 hours, the cell proliferation was assayed by CCK-8. The cell cycle was assayed by flow cytometry, the cells were transplanted to nude mice to establish the model, the apoptosis index was assayed by Tmmel, the positive expression index of ki-67 was assayed by concentrations of capsaicin on the ECI09 cells could inhibit cell proliferation and disturb the cell cycle, increase the number of cells in G0-G1 phase in a manner. In vivo experiment, the expression ofki-67 was detected by the method of Tunnel and immunohistochemistry in each group and indicated that the capsaicin could inhibit cell proliferation and promote apoptosis. Conclusion The different concenlrations of capsaicin can inhibit cell proliferation and promote apoptosis in a concentration-dependant manner on the EC 109 cells of esophageal squamous cell carcinoma.
