摘 要:目的利用高效液相色谱法同时测定黑参中4种稀有皂苷、18种氨基酸,紫外分光光度法测定黑参多糖、多酚的含量,研究不同产地的黑参中皂苷、氨基酸、多糖和多酚成分的差异,为黑参质量标准的建立奠定基础。方法 (1)采用COSMOSIL C18-PAQ色谱柱(ID 4.6 mm×250 mm,5μm),以水-乙腈为流动相进行梯度洗脱,检测波长203nm;柱温30℃,流速1.0 ml/min,进样体积10μl,测定4个不同产地黑参中4种稀有皂苷含量。(2)采用Waters Symmetry C18色谱柱(4.6 mm×150 mm,5μm),以水-乙腈为流动相进行梯度洗脱,检测波长254 nm;柱温40℃,流速ml/min,进样体积10μl,测定黑参中氨基酸含量。(3)分别以葡萄糖、没食子酸为对照,用浓硫酸-苯酚法测定黑参多糖及Folin酚还原法测定黑参多酚含量。结果不同产地黑参化学成分的含量有一定差异,如多糖含量高低之间相差3倍,多酚含量高低相差2.7倍,人参皂苷单体Rk1高低之间相差12.3倍,18种游离氨基酸总量相差2.1倍。结论首次同时对4地区黑参的人参皂苷、游离氨基酸、多糖及多酚进行了含量测定,为黑参质量标准的建立提供一定依据。Objective To determine the contents of saponins, amino acids, polysaccharides and polyphenols in black ginsengs by HPLC and UV method. In the meantime, the difference of the above components in black ginsengs from different areas was compared. Methods (1) COSMOSIL C18-PAQ (4.6 IDx250 mm, 5 μm) column was used to determine the contents of 4 kinds of rare saponins of black ginsengs from 4 different habitats. Acetonitrile water gradient was eluted as mobile phase The wavelength for measurement was 203 nm, with a flow rate of 1.0 ml/minat 30℃, and sample volume was 10 gl. (2) Waters Symmetry C18 (4.6x 150 mm, 5 μm) column was used to determine amino acid content in black ginseng. The mobile phase was the same as above. The wavelength for measurement was 254 nm, with a flow rate of 1.0 ml/min at 40℃, and sample volume was 10 gl. (3) Using glucose and gallic acid as the control, the polysaccharides content of black ginsengs was determined by the method of concentrated sulfuric acid and phenol. The polyphenols content was determined by Folin phenol method. Results The active ingredients of the black ginsengs from different places were different. The difference between the highest and lowest in the contents of saponin monomer Rkl, total amount of 18 free amino acids, polysaccharide, and polyphenol were 12.3, 2.1, 3, 2.7 folds, respectively. Conclusion The comtents of saponins, amino acids, polysaccharides and polyphenols in black ginsengs are determined, which can provide a basis for the establishment for quality standard for black ginsengs.
