摘 要:研究通过比较5种方法提取花色苷,发现乙醇/硫酸铵双水相体系提取黑豆皮中花色苷得率最高。通过单因素试验确定双水相提取的最优条件,采用响应面法优化了双水相法提取黑豆皮花色苷工艺。结果表明:双水相法提取黑豆皮花色苷最佳工艺为料液比1∶56(g/mL),硫酸铵质量分数22%,乙醇质量分数30%,pH3.0,花色苷得率为2.81 mg/g。此研究为花色苷实际生产应用提供理论依据。Comparing five methods to extract anthocyanins, it was found that the extraction rate of anthocyanins in black bean hulls was the highest by ethanol/ammonium sulfate aqueous two phase system.The optimum con-ditions for the extraction of aqueous two phase were determined by single factor test. The response surface methodology was used to optimize the extraction of anthocyanins from black soybean hull by aqueous two phase. The optimal parameters were optimized by response surface analysis. The liquid ratio, ammonium sulfate mass fraction, ethanol concentration, pH were 1:56 (g/mL), 22%, 30%, 3.0 respectively. The optimal yield of anthocyanin was 2.81 mg/g. The research will provide theoretical support for the anthocyanin of practical appli-cation.
