摘 要:目的获得高产的工业微生物产生菌。方法采用ARTP诱变的方式来获得头孢菌素C的高产突变株。通过对突变菌株采用混合培养筛选与高通量生物检测相结合的高效筛选方法。结果最终选育获得高产菌株G6。与出发菌株W-6相比,G6在摇瓶中产头孢菌素C的能力提高了19.75%。结论 ARTP诱变方法是一种高效的头孢菌素C高产菌株选育方法。其正突变几率高,可有效提高菌株的头孢菌素C产量。Objective To improve microbial strains for the high-production of industrial products.Methods In this paper,in order to obtain a cephalosporin C (CPC) high-yield stain,atmospheric and room temperature plasma (ARTP) was employed to generatethe mutants.In this study,all the mutants were given the equal screening chance by a mixture culture method combined with a high-throughput screening bioassay.Results The high-yield mixture strain G6 was successfully screened out and the CPC titer was nearly 19.75%,higher than that of the parental strain W-6 in the shake flask.Conclusion This work provides an appropriate strategy for obtaining high CPC-yield strains by using ARTP random mutation methods with an efficient screening assay.The ARTP random mutation method has a high positive mutation rate and the yield of cephalosporin C is significantly increased.
