摘　　要：目的:研究Notch1在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)组织中的表达变化情况,并探讨其相关临床意义。方法:采用免疫组化、实时荧光定量PCR技术检测52例肾透明细胞癌、40例癌旁肾组织中Notch1蛋白和mRNA的表达。结果:肾透明细胞癌及癌旁组织中Notch1蛋白阳性表达率分别为80.8%(42/52)、30.0%(12/40)(P<0.05);Notch1蛋白表达与肾透明细胞癌的TNM分期、Fuhrman分级、远处转移有关(P<0.05);肾透明细胞癌及癌旁组织中Notch1 mRNA相对表达量分别为0.317±0.044、0.153±0.038(P<0.05)。结论:Notch1蛋白及mRNA在肾透明细胞癌组织中表达明显增高。但在肾透明细胞癌发生、发展过程中的作用仍有待探讨。Objective：To explore mRNA expression and significance of Notchl in clear cell renal cell carcinoma （ccRCC）. Methods： Immunohistochemistry,real - time fluorescent quantitative PCR technique were selected to de-tect Notchl proteins and mRNA in 52 cases of ccRCC,40 cases of renal tissue adjacent to carcinoma. Results： Notchl protein positive expression rate in the ccRCC and the tissue adjacent to carcinoma were 80. 8% （42/52 ） , 30. 0%（12/40） （ P 〈 0. 05 ） . Notchl protein expression was correlated with TNM stage, grade of Fuhrman and distant metas-tasis （P〈0.05）. Notchl mRNA relative expression in ccRCC and the tissue adjacent to carcinoma were 0.317 ± 0. 044,0. 153 ± 0. 038 （ P 〈 0. 05 ）. Conclusion： Notchl is highly expressed in the ccRCC. Notchl may play a vital role in the development of the ccRCC,either promoting or inhibiting the carcinogenesis process.
