摘 要:目的建立应用高效液相色谱法测定保健品中叶黄素的分析方法。方法样品经无水乙醇分散溶解、碱液皂化、有机溶剂萃取后,分取适量氮吹浓缩,以0.1%BHT乙醇溶液溶解定容,以甲醇为流动相,流速为1.5 mL/min,柱温30℃,经由YMC Carotenoid色谱柱分离,采用高效液相色谱-光电二极管阵列检测器在445nm下进行检测,以峰面积外标法定量。结果加标回收率实验中,3个添加浓度水平下,叶黄素的平均回收率在94.7%~101%之间,相对标准偏差为0.87%~3.97%。方法检出限为1.5 mg/kg、定量限为5.0 mg/kg。结论该方法具有操作简便、快速、准确的特点,适用于保健品中叶黄素的含量分析。Objective To establish a method for determination of lutein in health products by high performance liquid chromatography(HPLC). Methods Samples were dissolved in ethanol, saponified by alkaline solution, extracted by organic solvent, using methanol as mobile phase, flow rate was 1.5 mL/min, column temperature was 30 ℃, separated by YMC Carotenoid column, detected by HPLC with photo-diode array detector(PDA) at 445 nm, and quantified by the external standard method. Results At different spiked levels, the average recoveries of lutein were between 94.7%~101%, with the RSDs of 0.87%~3.97%. Limit of detection and limit of quantitation were 1.5 mg/kg and 5.0 mg/kg, respectively. Conclusion The method is simple, rapid and accurate, which can be applied to the analysis of the content of lutein in health products.
