摘  要:目的研究表没食子儿茶素没食子酸酯(EGCG)对人口腔鳞状细胞癌细胞株Tca8113细胞C-myc、PTEN基因表达的影响。方法 MTT检测EGCG对Tca8113细胞增殖的抑制作用;荧光显微镜观察细胞形态的差异;real-time PCR检测C-myc、PTEN基因的表达。结果 MTT检测显示EGCG明显抑制Tca8113细胞的增殖;荧光显微镜显示研究组细胞中出现了大量的凋亡细胞;real-time PCR结果显示Cmyc基因表达降低,PTEN基因表达升高。结论 EGCG抑制了Tca8113细胞的生长,并改变了原癌基因C-myc、抑癌基因PTEN的表达。Objective To investigate the effects of epigallocatechin gallate( EGCG) on the expression of C-myc and PTEN in human oral squamous cell carcinoma cell line Tca8113. Methods MTT was used to detect the inhibitory effect of EGCG on the proliferation of Tca8113 cells. The morphology of cells was observed by fluorescence microscope. The expression of C-myc and PTEN was detected by real-time PCR. Results MTT assay showed that EGCG significantly inhibited Tca8113 cell proliferation; fluorescence microscopy showed that there were a lot of apoptosis cells in the experimental group; real-time PCR showed reduced expression of C-myc gene,PTEN gene expression increased.Conclusions EGCG inhibited the growth of Tca8113 cells,and altered the expression of proto oncogene C-myc and tumor suppressor gene PTEN.
