Optimization the Flask-shaking Fermentation Conditions of an L-isoleucine Producing Strain Brevibacterium flavum by Response Surface Method

摘  要:对黄色短杆菌突变株BM2610生产L-异亮氨酸的摇瓶发酵条件进行了研究.单因素优化了菌株BM2610的发酵培养基与培养条件,确定了发酵培养基的最佳碳氮源、最适起始pH、装液量及接种量等,进一步通过响应面分析方法优化了发酵培养基的组成配比.结果在优化条件下,BM2610摇瓶发酵平均积累L-异亮氨酸16.11 g·L^(-1),比未优化前提高了126.3%.The flask-shaking batch fermentation conditions of mutant strain Brevibacterium flavumBM2610 were studied. The single factor for fermentation medium and culture conditions for pro- ducing L-isoleucine by strain BM2610 were optimized, respectively. The optimum carbon and nitrogen source of fermentation medium, the optimized initial pH, volume of broth and inoculation amount for fermentation medium were determined. The composition and ratio of fermentation medium were further optimized by response surface method. Under the optimum fermentation condition, the L-isoleucine production of B.flavum BM2610 reached to 16. 11 g · L^-1 by flask-shaking ferinentation, which was increased 126.3 % compared to that under the initial fermentation condition.
