摘  要:目的探讨miR-7-5p是否通过调控EGFR的表达而影响人非小细胞肺癌细胞体外增殖和侵袭能力。方法通过双荧光素酶报告证明EGFR为miR-7-5p的下游靶基因,miR-7-5p及其阴性对照(miR-N.C.)转染人非小细胞肺癌细胞株NCI-H1975,观察细胞中EGFR的mRNA水平和蛋白水平的表达变化;然后通过MTS细胞增殖实验、细胞周期实验和Trans-well侵袭实验,观察NCI-H1975细胞体外增殖和侵袭能力的变化。结果与对照组相比,miR-7-5p可显著降低荧光素酶的表达活性(70.6%vs 100%,χ~2=28.3,P<0.01)。miR-7-5p靶向作用EGFR可使NCI-H1975细胞中mRNA表达水平较对照下调78.9%(χ~2=114.9,P<0.05),而蛋白水平则下调了71.4%(χ~2=113.3,P<0.05)。miR-7-5p处理后第3天,NCI-H1975细胞的增殖能力比对照组细胞降低了32.6%(χ~2=27.1,P<0.05)。而细胞周期实验表明与阴性对照相比,miR-7-5p处理组的NCI-H1975细胞,其S期所占比例显著下调(19.2%vs 32.7%,χ~2=5.2,P<0.05),而G0/G1期所占比例则显著增加(67.8%vs 55.1%,χ~2=2.9,P<0.05)。相比对照组,miR-7-5p可将NCI-H1975细胞的侵袭能力下调约47.3%(χ~2=48.9,P<0.05)。结论 MiR-7-5主要是通过靶向抑制EGFR表达减弱人非小细胞肺癌的增殖和侵袭能力。Objective To investigate the effect of miR-7-5 p on the proliferation and invasion of human nonsmall cell lung cancer cells by regulating the expression of EGFR. Methods EGFR was a downstream target gene of miR-7-5 p by dual luciferase reporter assay. Human non-small cell lung cancer cell line NCI-H1975 was transfected with miR-7-5 p and negative control( miR-N. C),then RT-RCR and western blot were used to detect the expression of EGFR in mRNA and protein level. It used MTS cell proliferation assay,cell cycle assay and Trans-well invasion assay to detect the proliferation and invasion of NCI-H1975 cells. Results Compared with the control group,miR-7-5 p significantly decreased the luciferase activity of EGFR( 70. 6% vs 100%,χ^2= 28. 3,P〈0. 01),and EGFR mRNA level and protein level decreased 78. 9% and 71. 4% in NCI-H1975 cells,respectively. On the third day after miR-7-5 p treatment,the proliferation of NCI-H1975 cells decreased about 32. 6%( χ^2= 27. 1,P〈0. 05) compared with the control group. The cell cycle experiments showed that the percentage of S decreased significantly( 19. 2% vs32. 7%,χ^2= 5. 2,P〈0. 05),and the proportion of G0/G1 phase increased significantly( 67. 8% vs 55. 1%,χ^2=2. 9,P〈0. 05) compared with the negative control group. miR-7-5 p inhibited the invasion ability of NCI-H1975 cells about 47. 3%. Conclusion MiR-7-5 suppresses the proliferation and invasion of human non-small cell lung cancer by inhibiting the expression of EGFR.
