摘  要:目的探讨TRAF2与Caspase-8在胃腺癌组织中的表达及其临床意义,并分析两者之间的相关性。方法分别采用qRT-PCR及Western blot法检测60例新鲜胃腺癌组织和55例癌旁正常胃黏膜组织中TRAF2、Caspase-8 mRNA及蛋白的表达。结果胃腺癌组织中TRAF2 mRNA和蛋白相对表达量均高于癌旁正常胃黏膜组织;胃腺癌组织中Caspase-8 mRNA及蛋白相对表达量均低于癌旁正常黏膜组织,差异均有统计学意义(P<0.05)。TRAF2和Caspase-8蛋白在胃腺癌组织中的表达呈负相关(P<0.05,r_s=-0.68),两者mRNA在胃腺癌组织中的表达无明显相关性。胃腺癌组织中TRAF2及Caspase-8蛋白表达均与患者性别、年龄无关(P>0.05);与分化程度、淋巴结转移、TNM分期及浸润深度有关(P<0.05)。结论 TRAF2因子在胃腺癌中表达上调,而Caspase-8表达下调,两者呈负相关,在胃腺癌中TRAF2因子可能通过下调Caspase-8发挥抗凋亡作用。联合检测TRAF2、Caspase-8有可能作为胃腺癌的预后指标,并有望成为胃腺癌治疗的新靶点。Purpose This paper aims to explore the expression of TRAF2 and Caspase-8 in gastric carcinoma and its clinical significance, and to analyze the relationship between them. Methods qRT-PCR and Western blot were used to detect the expression of TRAF2, Caspase-8 mRNA and protein in60 cases of fresh gastric adenocarcinoma and 55 cases of normal mucosa adjacent to cancer. Results Expression of TRAF2 mRNA and protein level in gastric carcinoma was higher than that of the adjacent normal gastric mucosa. While relative expression of Caspase-8 mRNA and protein in gastric carcinoma was lower than that of the adjacent normal mucosa tissue. The differences were statistically significant( P〈0. 05). In gastric cancer tissues, TRAF2 protein expression was negatively correlated with Caspase-8 protein expression(P〈0. 05, rs =-0. 68), but the correlation between mRNA in gastric adenocarcinoma tissue analysis was not statistically significant. Correlations were found between TRAF2, Caspase-8 protein expression and differentiation grade, lymph node metastasis, TNM staging and invasion depth of gastric cancer( P〈0. 05). Conclusion The expression of TRAF2 is up-regulated in gastric adenocarcinoma, while the down-regulation of Caspase-8. There is negative correlation between them. In gastric adenocarcinoma, TRAF2 factor may play an anti-apoptotic role by down regulating Caspase-8. TRAF2 and Caspase-8 are closely related to the clinicopathological features of gastric adenocarcinoma,which may be a prognostic marker of gastric adenocarcinoma and a new target for the treatment of adenocarcinoma of the stomach.
