摘 要:植物根际促生菌因其能够促进植物生长、防治病害的优势日益受到广泛关注,其研究主要集中在假单胞菌类,假单胞菌类在植物根际的定殖主要与趋化作用相关。本研究通过生物信息学软件对恶臭假单胞菌UW4的甲基趋化受体蛋白(MCPs)进行筛选,然后对其进行克隆和表达,结果表明:克隆得到的MCP1可稳定表达,通过对恶臭假单胞菌UW4的甲基趋化受体蛋白的纯化及分析其与不同配体物质的热力学作用强弱,以期找出恶臭假单胞菌特异性的趋化受体,进一步阐明其趋化机理,为恶臭假单胞菌对植物的促生效应提供理论支撑。The plant growth-promoting rhizosphere bacteria became more and more striking because it could promote the growth, prevent and treat diseases of plant. These researches mainly concentrated on the Pesudomonas. Pesudomonas colonization in plant rhizosphere was related to chemotaxis. This study screened the methyl chemotaxis receptor proteins(MCPs) of Pesudomonas UW4 by using several bioinformatics software, and then cloned and expressed the MCPs. The result showed that the cloned MCP1 could express stably, through the purification of the methyl chemotaxis receptor proteins of Pseudomonas UW4 and analysis the thermodynamic effect of the different ligands with MCPs, we could find out the specificity chemotaxis receptors of Pesudomonas UW4, clarify its chemotaxis mechanism and give some theoretical supporting to the growth-promoting of plant.
