摘 要:利用生物信息学方法对β-葡萄糖苷酶Bgl2238的氨基酸序列进行分析。采用Prot Param、DNAMAN、Multicoil、SOPMA、PROSAN等软件对其理化性质、亲/疏水性、不稳定指数、PEST序列、蛋白质二级结构及蛋白质修饰位点等重要参数进行预测,并对其三级结构进行同源建模及模建结果质量的评估。结果发现:Bgl2238由745个氨基酸组成,蛋白质序列中α-螺旋占36.64%,β-延伸链占19.06%,β-转角占9.40%,无规则卷曲占34.90%,存在14段非PEST序列,具有41个不同蛋白质修饰位点。采用在线网站SWISS-MODEL建模得到了Bgl2238的三级结构,分析蛋白质可及性并分析得到Ramachandran图,检测结果说明三维结构是合理的。本研究结果为β-葡萄糖苷酶Bgl2238进一步研究水解功能及提高其酶活性奠定了理论基础。The amino acid sequence of β-glucosidase Bgl2238 was analyzed by bioinformatics. The important parameters, such as physicochemical properties, hydrophobicity/hydrophilicity, instability index, PEST sequence,protein secondary structure and protein modification sites were predicted by using Prot Param, DNAMAN,Multicoil, SOPMA, PROSAN and other softwares. The homologous modeling for its tertiary structure and the evaluation on the quality of the modeling results were also carried out. The results showed that Bgl2238 was composed of 745 amino acids. The protein sequence comprised 36.64% of α-helix, 19.06% of β-extensionchian,9.40% of β-turn and 34.90% of atactic coil. There were 14 non-PEST sequences and 41 different protein modification sites. The protein tertiary structure of Bgl2238 was obtained by SWISS-MODEL modeling. The accessibility of the protein was analyzed and the Ramachandran graph was obtained. The results of this research showed that the three-dimensional structure was reasonable, which might provide a theoretical foundation for further study on the hydrolysis function of β-glucosidase Bgl2238 and the enhancement of its enzymatic activity.
