摘 要:目的探讨玉米须多糖对糖尿病小鼠模型的护肝降糖作用及其对肝糖代谢的影响。方法将50只小鼠分为空白对照组(NC组)、高血糖对照组(HG组)、二甲双胍组(MET组)、25 mg/kg玉米须多糖组(SMP25组)及50 mg/kg玉米须多糖组(SM P50组),每组10只。采用腹腔注射四氧嘧啶方法制备糖尿病小鼠模型,利用血糖测试仪测定小鼠空腹血糖(FPG)浓度,通过比色法测定小鼠肝糖原含量,通过药物性肝损伤病理组织学评分系统(DILI-PSS)评估肝损伤情况。结果与HG组相比,SMP50组小鼠的FPG降低34.3%(P<0.001)、肝糖原含量提高321.5%(P<0.001);与MET组相比,SMP25、SMP50组小鼠肝糖元含量均升高(P<0.001),且肝细胞水肿和脂肪性变均减轻(P<0.001);SMP50组的降糖护肝作用优于SMP25组(P<0.001)。结论玉米须多糖可降低糖尿病小鼠的FPG,促进肝糖元的合成,并可减轻造模药物及高血糖引起的肝损害,有望成为糖尿病患者护肝降糖辅助治疗选择。Objective To investigate the effects of stigma maydis polysaccharides on glycometabolism and liver protection in diabetic mice. Methods Fifty mice were divided into normal control group(NC group),hyperglycemia control group(HG group),metformin group(M ET group),25 mg/kg polysaccharide derived from stigma maydis group(SMP25 group),and 50 mg/kg polysaccharide derived from stigma maydis group(SMP50 group),ten in each group.The diabetic mice models were prepared by peritoneal injection of alloxan,the fasting plasma glucose(FPG) were measured by a blood glucose tester,the content of liver glycogen in mice were measured by colorimetry,and the liver injury was evaluated by drug-induced liver injury-pathological scroring system(DILI-PSS). Results Compared withHG group,FPG value was reduced by 34.3%(P〈0.001),the content of glycogen was increased by 321.5% in SM P50 group; compared with M ET group,the contents of glycogen were increased(P〈0.001),and hepatocyte edema and hepatic steatosis were reduced(P〈0.001) in SM P25 and SM P50 groups; the effects of blood sugar control and liver protection in SM P50 group is better than those in SM P25 group(P 0. 001). Conclusion Polysaccharides derived from stigma maydis can significantly reduce the FPG concentration,promote the synthesis of glycogen,reduce the liver injury caused by modeling drugs and hyperglycemia in diabetic mice,and is expected to provide a effective adjuvant treatment option for diabetics.
