摘  要:本文以Methylobacterium sp.XJLW为出发菌株,从培养基选择、甲醇浓度、硫酸铵浓度、初始pH、发酵温度、渗透压和氧载体角度对其液态发酵合成PHB的工艺进行了初步研究,结果显示,当选择M3培养基,并将甲醇浓度调整为1.5%(v/v)、将硫酸铵浓度调整为0.5 g/L、初始pH=7.0、添加0.5%(v/v)Tween-80时,于30℃发酵5 d,菌体生物量可达到2.59±0.22 g/L,PHB含量达到737.63±44.61 mg/L,与初始M3培养基相比分别提高了35.6%、40.1%。这些数据表明Methylobacterium sp.XJLW可以甲醇为唯一碳源合成PHB,对于利用代粮发酵工艺合成PHB的研发具有重要参考意义。Methylobacterium sp. XJLW was used as original strain to produce PHB by submerged fermentation process. In order to improve the PHB productivity, the effects on medium type, concentration of methanol and ammonium sulfate, initial pH, fermentation temperature, osmotic pressure and oxygen vector on cell growth and PHB production were studied. Results showed that M3 medium was a superior design, the optimal methanol concentration was 1.5 %(v/v), the optimal ammonium sulfate concentration was 0.5 g/L, the optimal initial pH and fermentation temperature were 7.0 and 30 ℃, respectively. Increasing osmotic pressure could do no help to improve PHB production, but when 0.5 %(v/v) was added, DCW and PHB content were both increased. Finally, under optimal fermentation conditions, DCW of Methylobacterium sp. XJLW and PHB content were 2.59±0.22 g/L and 737.63±44.61 mg/L, which were 35.6 % and 40.1 % higher than the level in M3 medium system. These data will provide an important reference for research of PHB production through submerged fermentation without grain consumption.
