摘 要:研究了23种常见谷物杂粮的70%乙醇提取物对黄嘌呤氧化酶(XOD)的体外抑制作用。在质量浓度200μg/m L条件下,17种醇提物对XOD有抑制效果,其中3种杂粮的抑制率超过50%。抑制效果较好的前4种杂粮依次是荞麦籽、白高粱、麦仁、薏仁米,它们的IC50分别是156.7,159.2,195.9,196.4μg/L。动力学分析表明,白高粱、荞麦籽醇提物的抑制类型为竞争抑制,抑制常数Ki分别为41.04μg/L和15.79μg/L;而麦仁、薏仁米醇提物的抑制类型为混合抑制,竞争抑制常数Ki分别为23.67μg/L和28.76μg/L,非竞争抑制常数Kis分别为875.65μg/L和566.375μg/L。Xanthine oxidase(XOD) catalyzes the oxidation of hypoxanthine to xanthine then to uric acid, which plays a crucial role in gout. Ethanol extracts from twenty-three kinds of cereal and grain were investigated for the inhibition on XOD activity in vitro. Among the 23 ethanol extracts, 17 showed inhibitory effects at 200 μg/m L and 4 extracts showed higher inhibition rate near or beyond 50%. While 16 extracts and 10 extracts performed the activity at 100 μg/m L and 50 μg/m L respectively, none had the inhibition activity greater than 50%. Four more active extracts were the ethanol extracts of buckwheat seed, white sorghum seed, wheat kernel, and barley rice with IC50 values of 156.7, 159.2,195.9 μg/m L and 196.4 μg/m L. Further dynamics analysis showed that the inhibition type of buckwheat extract and white sorghum seed extract on XOD were competitive inhibition, the inhibition constant(Ki) were obtained as 41.04 μg/m L and15.79 μg/m L. However, the inhibition type of wheat kernel extract and barley rice extract were mixed inhibition. The competitive inhibition constant(Ki) were 33.36 μg/m L and 23.67 μg/m L when the noncompetitive inhibition constant(Kis)were 747.28 μg/m L and 875.65 μg/m L.
