摘 要:目的利用海洋芽孢杆菌SCSIO06063来源的蛋白酶对扇贝蛋白进行酶解制备抗氧化肽。方法采用超滤和反复色谱分离纯化扇贝水解物。获得的肽类结构通过基质辅助激光解析飞行时间质谱(MALDI-TOF-MS/MS)鉴定,采用DPPH自由基清除实验评价肽类的抗氧化活性。结果分离纯化后获得3个肽类组分A、C和E,依次被鉴定为His-Ala-Ile-Ile-Met-Met-Val(A)、Leu-Asn-Pro-Lys-LeuIle-Met-Val(C)和Met-Ile-Leu-Ile-Leu-Phe-Arg(E),经NCBInr数据库中Blast检索,未发现A组分的同源序列,提示其为一个新的肽类结构。A、C和E 3个组分均具有一定的抗氧化能力,在浓度为1.0mg·m L-1时,它们的清除率分别为51.6%、55.4%和53.4%。结论海洋芽孢杆菌SCSIO06063来源的蛋白酶可用于扇贝蛋白酶解制备抗氧化肽。Objective To prepare antioxidant peptides with proteinase from Bacillus sp. SCSIO 06063 (P-SCSIO 06063). Methods Ultrafiltration and repetitive chromatographic methods were used to separate and purify the hydrolysate of scallops protein. The sequences of peptides were identified with matrix assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS/MS). The antioxidant activity of these peptides was tested with 1, 1-diphenyl-2-picrylhydrazyl (DPPH) scavenging assay. Results Three peptides were obtained and identified as His-Ala-Ile-Ile-Met-Met-Val (A), Leu-Asn-Pro-Lys-Leu-Ile-Met-Val (C), and Met- Ile-Leu-Ile-Leu-Phe-Arg (E) by MALDI-TOF-MS/MS. These peptides were compared to the NCBInr database by Blast, which showed that A was not similar to any bioactive peptides, indicating it was a new peptide. A, C, and E at 1.0 mg· mL-1 showed DPPH scavenging rate of 51.6%, 55.4%, and 53.4%, respectively. Conclusion The proteinase from Bacillus sp. SCSIO06063 can be used in the preparation of antioxidant peptides.
