摘  要:目的:探讨BIRC5基因rs2071214位点与缺血性脑卒中发病的关联性。方法:采用高分辨率熔解曲线(HRM)技术对513例缺血性脑卒中患者和515例健康对照者的BIRC5基因rs2071214位点进行基因分型,并对比两组之间的基因型和等位基因的分布频率以及两组之间一般临床资料和生化指标的差异。选取80例病例和80例对照,运用实时荧光定量PCR技术检测BIRC5基因在两组之间的mRNA表达水平。结果:病例组和对照组BIRC5基因rs2071214位点基因型分布无统计学差异(P〉0.05),等位基因A、G的分布频率在两组之间有统计学差异(P=0.041)。比较两组之间的显性模型(AAvs AG+GG)和隐性模型(GGvs AG+GG)的基因型分布,隐性模型中两组之间的差异有统计学意义(P=0.046)。病例组BIRC5基因的mRNA表达水平略高于对照组,但两组之间的差异无统计学意义(P〉0.05)。结论:BIRC5基因rs2071214位点与缺血性脑卒中有一定的相关性。Objective:To investigate the association between ischemic stroke(IS)and BIRC5 rs2071214 polymorphism.Methods:High resolution melting(HRM)was used to detect BIRC5 rs2071214 polymorphism in 513 patients with IS and 515 healthy controls.The genotype,allele frequency,clinical data and biochemical index were compared between the two groups.Real time fluorescent quantitative PCR(RT-qPCR)was used to test the mRNA expression of BIRC5 in 80 patients with IS and 80 controls.Results:There was no significant difference(P〈0.05)in genotype distribution of BIRC5 rs2071214 between the cases and controls.The distribution of allele A and allele G was statistically different(P=0.041)between the two groups.There was a statistical difference(P=0.046)in recessive model after comparing the genotype distribution of dominant model(AA vs AG+GG)and recessive model(GG vs AG+AA)between the two groups.The mRNA expression of BIRC5 in cases was higher than in controls,and no statistical difference was found(P〈0.05).Conclusion:The BIRC5 rs2071214 polymorphism might be related to ischemic stroke.
