不同PUFAs调节脂肪细胞脂联素和PPAR γ的剂量反应关系

摘  要:目的探讨不同类型多不饱和脂肪酸(PUFAs)调节3T3-L1脂肪细胞脂联素及过氧化物酶体增殖物激活受体γ(PPARγ)的剂量反应关系和效应差异。方法不同浓度(0、25、50、100、200、400μmol/L)二十碳五烯酸(EPA)、二十二碳六烯酸(DHA)、α-亚麻酸(α-ALA)、花生四烯酸(AA)及亚油酸(LA)处理3T3-L1脂肪细胞24h,实时荧光定量PCR法测定脂联素及PPARγ基因表达水平,Western blot法测定二者蛋白水平,ELISA法测定脂联素分泌水平。结果 50~200μmol/L浓度范围内,n-3PUFAs上调脂联素及PPARγ基因表达,DHA作用最显著(P<0.05);400μmol/L呈现显著抑制效应。100μmol/L时,n-3 PUFAs上调脂联素及PPARγ蛋白表达(P<0.05);仅DHA促进脂联素分泌(P<0.05),n-6PUFAs呈现抑制效应。结论 PUFAs在一定浓度范围内提升脂联素及PPARγ基因表达,高浓度作用时呈现抑制效应。n-3PUFAs更能促进脂联素、PPARγ蛋白合成,并促进脂联素分泌。不同类型PUFAs对PPARγ的不同作用,可能是其对脂联素的调节产生差异的重要原因。Objective To explore dose-dependent effects of polyunsaturated fatty acids(PUFAs) on adiponectin and peroxisome proliferator-activated receptor-γ(PPARγ) expression in 3 T3-L1 adipocytes. Methods 3 T3-L1 adipocytes were incubated with eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA), α-linolenic acid(α-ALA), arachidonic acid(AA) and linoleic acid(LA) at 0,25, 50, 100, 200, 400 μmol/L. The m RNA levels of adiponectin and PPARγ in adipocytes were determined by real-time PCR. Cellular protein levels were determined by Western blot. Adiponectin levels in media were determined by ELISA. Results N-3 PUFAs increased m RNA levels of adiponectin and PPARγ at 50-200 μmol/L, while the most significant increase of adiponectin was induced by DHA(P〈0.05). PUFAs treatments at 400 μmol/L showed inhibitory effects. At 100μmol/L, n-3 PUFAs increased the syntheses of adiponectin and PPARγ(P〈0.05). DHA promoted the secretion of adiponectin, while n-6 PUFAs showed inhibitory effects(P〈0.05). Conclusion PUFAs can increase m RNA levels of adiponectin and PPARγ at certain concentrations, while high concentrations show inhibitory effects. n-3 PUFAs more effective in inducing the syntheses of adiponectin and PPARγ, and promoting the secretion of adiponectin. The effects of different PUFAs on PPARγ may contribute to their distinct regulatory effects on adiponectin.
