摘 要:为了分析2008-2011年北京港口进出口水产品中非O1/O139群霍乱弧菌的多位点序列分型(MLST)特征,为霍乱弧菌导致食品安全事件的有效溯源提供理论支持,选取2008-2011年北京港口食源性非O1/O139群霍乱弧菌分离株67株,采用毒力基因PCR检测并用MLST分析。结果:毒力基因PCR检测结果显示,所有菌株均未检出CTX、tox R、hly A 3种基因。菌株的MLST结果显示,菌株可被分为62个ST型,具有明显的多样性。其中原有STs 2个,新发现STs 60个。同ST型菌株均来源于同一国家,本实验数据同pubmlst数据库中数据进行分析,有7个STs可以归为7个克隆组,它们来源于中国和泰国,同组内STs来源于中国、澳洲、法国、泰国和非洲。表明北京港口非O1/O139群霍乱弧菌遗传特征复杂多样,MLST方法可作为可靠的食品污染溯源手段,相同地区的菌株有一定的地域特点,此研究丰富了现有霍乱弧菌MLST数据库,为其溯源提供支持。Multilocus sequence typing( MLST) was ued to analyze non-O1/O139 Vibrio cholerae( V. cholerae) to provide theoretical support for effective traceability in food safety incidents caused by foodborne V. cholerae,which was isolated from import and export food from Beijing port between 2008 to 2011. Methods: Totally of 67 non-O1/O139 V. cholerae strains were isolated from food at Beijing port between 2008 to 2011. The virulence gene were determined by PCR,and molecular typing was carried out by multilocus sequence typing( MLST). Results: PCR detection of virulence gene showed that all the strains were negative for CTX,tox R,and hly A3. MLST molecular typing results showed that the V. cholerae was divided into 62 ST types,2 STs exised in pubmlst database and 60 STs were new discoveries. Same STs were all derived from the same country. After analyzing with other Pubmlst database,we found that 7 new STs were grouped into 7 exist groups which were from China and Thailand. Others within the group are were from China,Australia,France,Thailand and Africa. Conclusion: The genetic characteristics of non-O1/O139 V. cholerae strains isolated from food at Beijing port are complex and have great diversity. The MLST method can be used as a reliable method to analyze the source of food contamination. The strains in the same area have certain geographical characteristics. This study enriches the exisiting MLST of V. cholerae database to support traceability.
